In a medium saucepan, combine the strawberries, sugar, and water. Bring to a boil over medium-high heat, stirring occasionally.
Reduce heat to low and simmer for 5 minutes, stirring occasionally. Remove from heat and stir in the vanilla extract.
Allow the mixture to cool completely.
In a medium bowl, combine the mascarpone cheese, heavy cream, and confectioners' sugar. Beat with an electric mixer on high speed until light and fluffy.
In a trifle bowl, layer half of the ladyfingers, half of the cooled strawberry mixture, and half of the mascarpone cream mixture. Repeat with remaining ingredients.
Drizzle the Kahlua over the top of the trifle.
Refrigerate for at least 2 hours before serving.
